Because the voltage-gated sodium channel Nr1 tetramer is a prominent target for metabotropic glutamate receptors (mGluR) in glioblastoma, we investigated the effects of mGluR ligands on the voltage-gated sodium channels Nav 1.2 and Nav1.3. U-69593, LY354740, and MPEP, three selective mGluR5 antagonists, decreased Nav1.2 and Nav1.3 expression and current density, whereas both LY341495, a selective mGluR4 antagonist, and AMG 362, an mGluR7 antagonist, had no effect. U-69593, LY354740, and MPEP decreased Nav1.2 and Nav1.3 current density at all potentials with a more pronounced effect at depolarized potentials. These findings suggest that activation of mGluR5 may have an effect on Na+ channel function in glioblastoma and that all three mGluR5 agonists decrease the density of functional Na+ channels. The endocannabinoid, 2-arachidonoyl glycerol, is a selective agonist of mGluR5. Although 2-arachidonoyl glycerol induced proliferation of normal human astrocytes in culture, it decreased the proliferation of astrocytes derived from patients with glioblastoma and astrocytomas.
